Stem Cell and Developmental Biology: (1+2)
Theory:
Introduction: Cellular potency, lineage commitment, cellular development, differentiation, dedifferentiation & trans differentiation, Cell cycle control, Immortal DNA strand hypothesis, Asymmetric cell division, telomerases in relevance to stem cell development and differentiation.
Somatic and Germ cell derived stem cells: Germ line stem cells and germline, derived pluripotent cells, Stem cell niche, epithelial stem cells, mesenchymal stem cells, neural stem cells, haematopoitic stem cells, cardiac stem cells, Cancer stem cells, Markers, molecular and evolutionary mechanisms addressing origin and maintenance of cancer stem cells.
Regulatory mechanisms in Embryonic and adult stem cells: Coreregulatory circuitry, DNAmethylation, histone modifications, histone modifiers, chromatin remodelers, RNA Pol II code, post transcriptional control of gene expression in ESC: role of miRNAs, LincRNAs and RNA binding proteins. Spatial organization of genome during ESC development and differentiation.
Basic concepts of development: Potency, commitment, specification (autonomous, regulative and syncytial), induction, competence, determination and differentiation, morphogenetic gradients, cell fate and cell lineages, genomic equivalence and the cytoplasmic determinants, imprinting.
Gametogenesis and Fertilization: Production and structure of gametes, cell surface molecules in sperm egg recognition in animals, acrosome reaction, fast and slow block to polyspermy, zygote formation.

Media and Reagents: Types of cell culture media for plants, animals and microbial cells; Ingredients of media; Physiochemical properties; Buffers; Oxygen; Osmolarity; Temperature; Balance salt solutions; Antibiotics, growth supplements; Conditioned media; Other cell culture reagents; Preparation and sterilization of cell culture media and other reagents.
Animal Cell Cultures Techniques: History of animal cell culture; Different tissue culture techniques; Types of primary culture; Chicken embryo fibroblast culture; Chicken liver and kidney culture; Secondary culture; Trypsinization; Cell separation; Continuous cell lines; Suspension culture; Organ culture; Behaviour of cells in culture conditions: division, growth pattern, metabolism of estimation of cell number; Development, Characterization and maintenance of cell lines, Cryopreservation; Commercial scale production of animal cells, stem cells and their application; Application of animal cell culture for in vitro testing of drugs; Testing of toxicity of environmental pollutants in cell culture.
Stem cell and progenitor cell assays: Purification of tissue specific stem cells and transplantations. Hematopoietic progenitor cell analyses such as flow cytometry, dynabeads, colony forming assays and in&vitro differentiation assays of lymphoid (BandT), myeloid lineages and other tissue lineages.
Brief overview: Development in Health and Disease including birth defects, Development and the Environment (biotic, abiotic, and symbiotic regulation), Development and Evolution (developmental mechanisms of evolutionary change). Cell cycle analysis and Synchronization of cultures, cancer studies using cell culture, production of hybridoma and monoclonal antibody production. Animal cloning, Therapeutic cloning, Tissue engineering, Knock out animals.
